Bub3’s function doesn’t stop at the checkpoint

چکیده
Fewer forks make faster progress J ust as a large number of forks can inhibit someone unaccustomed to formal dining, an excess of replication forks can slow the rate of DNA synthesis, Zhong et al. reveal. Cdc7 is the catalytic subunit of the Dbf4-dependent kinase, which initiates DNA replication by activating the DNA helicase complex MCM at replication origins. In response to DNA damage, the checkpoint protein Rad53 limits origin fi ring by inhibiting Cdc7. Rad53 also slows the progression of existing replication forks, but whether Cdc7 infl uences fork dynamics is unclear. Zhong et al. found that, although cdc7 mutant yeast fi red fewer origins than wild-type cells, their replication forks proceeded faster than normal from the origins that did initiate DNA synthesis, even in the presence of the DNA-damaging agent MMS. orc1 mutant yeast, which fail to recruit MCM to replication origins, also showed faster fork progression at the handful of origins that managed to fi re, indicating that reduced levels of origin fi ring increase replication fork speed. mec1 mutants, on the other hand, initiated synthesis at more origins than wild-type cells and consequently showed decreased rates of replication fork progression. Because there are fewer replication forks in yeast lacking Cdc7 or Orc1, MMS only weakly activated the DNA damage checkpoint and Rad53 in these cells, suggesting that reduced checkpoint activation is one reason these cells show rapid fork dynamics. But mec1 mutants have slow replication forks even though they lack the DNA damage checkpoint. This suggests that the amount of origin fi ring infl uences fork progression in other ways, too, possibly because individual forks must compete for limited amounts of essential replication factors or nucleotides. Bub3 promotes Cdc20-dependent activation of the APC/C in S. cerevisiae
The spindle checkpoint ensures accurate chromosome segregation by sending a signal from an unattached kinetochore to inhibit anaphase onset. Numerous studies have described the role of Bub3 in checkpoint activation, but less is known about its functions apart from the spindle checkpoint. In this paper, we demonstrate that Bub3 has an unexpected role promoting metaphase progression in budding ye...
متن کاملTwo functionally distinct kinetochore pools of BubR1 ensure accurate chromosome segregation
The BubR1/Bub3 complex is an important regulator of chromosome segregation as it facilitates proper kinetochore-microtubule interactions and is also an essential component of the spindle assembly checkpoint (SAC). Whether BubR1/Bub3 localization to kinetochores in human cells stimulates SAC signalling or only contributes to kinetochore-microtubule interactions is debated. Here we show that two ...
متن کاملSuppression of a conditional mutation in alpha-tubulin by overexpression of two checkpoint genes.
To identify proteins that regulate microtubule assembly in Saccharomyces cerevisiae, we screened for multicopy suppressors of a conditional mutation in alpha-tubulin. Cells expressing the recessive allele tub1-729 as their sole alpha-tubulin gene grow normally at permissive temperature. However, at 15 degrees C the cells lose viability and arrest primarily with large buds and quantitatively dim...
متن کاملRecognizing chromosomes in trouble: association of the spindle checkpoint protein Bub3p with altered kinetochores and a unique defective centromere.
Spindle checkpoint proteins monitor the interaction of the spindle apparatus with the kinetochores, halting anaphase even if the microtubule attachment of only a single chromosome is altered. In this study, we show that Bub3p of Saccharomyces cerevisiae, an evolutionarily conserved spindle checkpoint protein, exhibits distinct interactions with an altered or defective kinetochore(s). We show fo...

The B-type cyclin Clb5 is involved primarily in control of DNA replication in Saccharomyces cerevisiae. We conducted a synthetic genetic array (SGA) analysis, testing for synthetic lethality between the clb5 deletion and a selected 87 deletions related to diverse aspects of cell cycle control based on GO annotations. Deletion of the spindle checkpoint genes BUB1 and BUB3 caused synthetic lethal...

Anaphase onset is regulated by the anaphase-promoting complex/cyclosome (APC/C), a ubiquitin ligase that when bound to the activator Cdc20 targets substrates for ubiquitination and subsequent degradation (London and Biggins, 2014b). If chromosomes are not attached to spindle microtubules in metaphase, the spindle checkpoint signals to inhibit APC/C substrate ubiquitination. Spindle checkpoint a...
